Purpose / Objectives 1. To develop an efficient technique for laboratory scale production of carrageenases (enzymes that degrade carrageena) and agarases (enzyme that degrade agar) using recombinant DNA techniques. Carrageenan and agar are red algae- specific, cell wall–localized polysaccharides. Enzymatic hydrolosis of these polysaccharides is critical to the preparation of protoplasts from red algae.
2. To develop and optimize techniques for preparation of protoplasts from Gracilaria and Kappaphycus and for fusion of the protoplasts to produce viable hybrids.
3. To develop genetic transformation techniques for species of Gracilaria.
