Purpose / Objectives To use Agrobacterium- mediated transformation of rice to:
a) evaluate the function of the rice homologue of the LEAFY COTYLEDON (LEC 1) gene and its ability to induce adventitious embryony in rice tissues when controlled by the rice actin1 promoter.
b) identify cis elements that are responsible for meiotic and mitotic expression of the rice homologue of the yeast DISRUPTED MEIOSIS cDNA1 (DMC1) gene.
Biosafety measures 1. The tested plants will be kept in isolation in the CL4 transgenic greenhouse to prevent access of unauthorized personnel.
2. Panicles of flowering plants will be bagged to prevent spread of pollen into environment.
3. Transgenic seeds will be harvested, stored, and analyzed for transgene integration and expression.
4. After analyses, the transgenic materials, pots and soil will be autoclaved and disposed as prescribed by NCBP.
