Purpose / Objectives 1. To use the pANDA vector to generate ~50 RNA interference (RNAi) lines for each selected drought-related genes.
2. To characterize the lines for alterations in carbohydrate and hormone metabolism, growth, cell division, fertility and drought tolerance
Biosafety measures 1. Transgenic plants (generations T0-T2) will be
kept in the CL4 transgenic greenhouse, where a security card system prevents access of unauthorized personnel
2. The plant materials, pots and soil for disposal from CL4 will be
placed in sealed plastic bags and autoclaved at hightemperature and high pressure
3. The plant material from screenhouse (CS07) will be disposed of by burning of straw
4. The access to the CS07 would be for authorized staff only.
5. A logbook will be maintained in CS07 to record the entry of personnel
6. CS07 will be kept locked
7. CS07 facility is adequate to prevent entry by rats, birds, etc.
8. Transgenic seeds harvested from CL4 and CS07 will be kept in hermetically sealed aluminum foil packages and stored in locked refrigerators designated for storage of transgenic seeds in TCGE Laboratory of IRRI. Movement of all materials will be done in compliance of all relevant biosafety and phytosanitary requirements of the Philippines.
