Purpose / Objectives • To undertake advancement by selfing of BC3F1 lines and produce BC3F2 seeds for multiloc trials.

• To generate local data for MAHYCO Event EE-1, which include:
a)whole plant assay to test the bioefficacy of MAHYCO event EE-1 against local population of eggplant fruit and shoot
b) effects on non-target arthropods;
c) comparative evaluation of horticultural and nutritional compositon.

• To generate baseline data on susceptibility of soil microfauna to Bt protein and mating behavior of the borer.
Biosafety measures • Pollen Flow within the Site-Bagging of flowers will be done prior to flower opening to prevent unintended transfer of pollen to non-transgenic plants. Any open flowers will be immediately removed.

•Pollen Flow outside the Site-The recommended isolation distance of at least 200 meters from other eggplant plantings will be followed (Agarwal, 1980 as cited by Lal, 1993; Chen, undated). Survey of the locality near the site indicates that there are no eggplant productions within 400 meters radius from the site.

•Limited Access to the Site-A perimeter fence was built to secure the area to prevent entry by unauthorized persons and stray animals. The gate is provided with a padlock and will be locked at all times. The site will be guarded 24 hours by a security guard to be hired by the project.

•Accidental Release to the Environment-Extreme care will be observed for any plant parts taken out of the confined trial site for sampling and further analysis. The samples will be placed in a closed container, labeled properly and transported by the most direct route to the designated facility. All sampling activities will be monitored by the members of concerned regulatory body. The plant parts will be chopped and disposed properly (boiled, autoclaved, buried) after use in designated areas to be approved by NCBP.

•Entry of Rodents/Rats-
The perimeter fence of the confined trial site consisted of the concrete posts surrounded by cyclone fence with barb wire at the lower and upper portion. A two-feet wide plain GI sheet was placed at the lower portion to prevent entry of rodents. As an added measure, a thick plastic sheet was placed below the GI sheet to ensure that the area will be rodent-free.
Conditions for Approval a)That all activities involving whole transgenic plants and viable plant materials be conducted only in the designated fenced facility, except for seeds that may be collected and stored in designated storage cabinets at the Institute of Plant Breeding, UPLB. Samples of plant parts may also be brought to the laboratory for analysis and/or molecular and biochemical characterizations. The designated and NCBP-approved fenced facility is the site located at the IPB Experiment Station, lots F-1 and F-2, Barangay Paciano Rizal, Bay, Laguna. The exact location of the area in 4 corner points as per GPS (Summit, Garmin) data is— point 1: 14? 08.653’ N, 121? 15.31’ E; point 2: 14? 08.681’ N, 121? 15.329’ E; point 3: 14? 08.664’ N, 121? 15.372’ E and point 4: 14? 08.637’ N, 121? 15.358’ E.

b)That all transgenic eggplants in the site contain the transformation event EE-1 only.

c)That no more than nine (9) transgenic DLP and ten (10) transgenic Mara lines be planted in the confined facility, according to the layout specified in the revised proposal.

d)That the proponents adhere as closely as possible to the planting and monitoring schedule.

e)That a temporal or physical reproductive isolation distance of at least 200 meters be maintained around the experimental site.

f)That modifications in the planting schedule and other activities could be made with the concurrence of the IBC and NCBP.

g)That Bt eggplants on the site be artificially infested with the eggplant fruit and shoot borer on or before DAS (days after sowing) 80.

h)That the detailed schedule of activities be revised to include additional Plant Quarantine Services, Bureau of Plant Industry (PQS-BPI) monitoring of activities for the following dates:
i.DAS 121 – Bagging of flowers for selfing, bagging and collection of flowers for quantitative ELISA, disease monitoring, arthropod sampling, EFSB damage monitoring.
ii.DAS 149 – Arthropod sampling, EFSB damage
monitoring, disease monitoring. Day(s) for harvesting of mature fruits. The revised detailed schedule of activities should reflect this activity and should also specify the frequency and duration of
harvesting mature fruits.

i)That an effective insect, rodent and fowl pest management scheme be put in place in the experimental area.

j)That the fenced test site be maintained as a limited access facility where only authorized personnel are allowed regular access during the implementation of the proposed activities.

k)That access to the fenced facility by guests, observers and other non-authorized personnel must have the prior approval of the NCBP and PQS-BPI. Requests for such access must be received by the NCBP and PQS-BPI at least five (5) days in advance.

l)That the NCBP and the PQS-BPI be informed immediately of any site intrusion by unauthorized persons.

m)That all plants and viable materials within the experiment area be accounted for.

n)That all plant materials taken out of the experiment area for analysis in the laboratory be disposed by heat treatment of plant parts followed by burial in a deep pit. That upon termination of the activities specified in the proposal and in the detailed schedule of activities, the plants will be uprooted, chopped and buried in the deep pit inside the site. The area should then be left undisturbed, but irrigated on a regular basis to induce germination of any seeds that might have remained in the ground after harvesting. The area will be observed for two (2) months after final harvesting. Any eggplant seedlings that would emerge should be uprooted, chopped and buried in a deep pit inside the fenced facility.

o)That the additional requirements that the NCBP may impose, as necessary, during the implementation of the experiment be complied with.
