Purpose / Objectives 1. To use biolistic transformation to increase the cytokinin level in the flagleaf of IR72 and new plant type (NPT) rice and thereby prolong photosynthesis during grain filling.
2. To transfer to rice a chimeric cytokinin biosynthesis gene in which the isopentenyl transferase (ipt) gene from Agrobacterium tumefaciens is controlled by senescenece- specific promoters (the SAG12 promoter from Arabidopsis and its rice homologue).
3. To transfer to rice a SAG12- gus construct to understand the regulation of SAG12 promoter function in rice.
4. To determine the effect of the SAG12 – ipt gene on cytokinin level in rice leaves.
5. To determine the effect of the gene on leaf senescence, endosperm development, yield and seed viability.
Biosafety measures 1. The transgenic rice produced will be grown in the Transgenic Greenhouse, which is a facility with containment capabilities.
2. The flowers of the plants will be bagged to prevent spread of pollen in the environment.
3. Seeds will be harvested, stored and analyzed for successful transformation and enhanced resistance to insects and diseases.
4. All other materials will be destroyed
by incineration.
