Purpose / Objectives 1. To produce transgenic rice with phytoene-synthase and phytoene desaturase which are major enzymes in â- carotene biosynthesis.
2. Transfer of the â-carotene genes from T3O9, japonica cultivar to indica cultivars by conventional breeding.
Biosafety measures 1. The tested plants will be kept in isolation in the CL4 transgenic greenhouse to prevent access of unauthorized personnel.
2. Panicle of flowering plants will be bagged to prevent release of pollen.
3. The plant materials for disposal will be placed in sealed plastic bags.
4. The plants used will be destroyed and the pots and soil will be autoclaved at high temperature and high pressure.
5. Transgenic seeds will be kept in hermetically sealed aluminum foil packages, inside a sealed metal container and stored in locked refrigerators designated for storage of transgenic seeds in Tissue Culture Laboratory of IRRI.
6. Transgenic T309 and control elite indica cultivars will be grown in CL4 greenhouse.
7. Homozygous transgenic T309 will be characterized.
8. Crossing with elite indica rice and
backcrossing will be continued until homozygous indica lines are
developed.
